Nicotinamide mononucleotide (NMN) supplementation increases blood NAD+ levels in healthy adults: a randomized, double blind, placebo controlled trial
npj Aging and Mechanisms of Disease, 2022
NAD+ is a coenzyme essential to energy production, DNA repair, and cellular regulation. Its levels decline by roughly 50% between young adulthood and middle age. NMN (nicotinamide mononucleotide) is the most direct NAD+ precursor available one biosynthetic step from NAD+, and the most studied route to raising NAD+ levels in human clinical research.
NMN is converted directly to NAD+ by a single enzyme (NMNAT), without the additional conversion steps required by niacin or nicotinamide. Human studies at 250 mg to 1,200 mg per day consistently show measurable increases in blood NAD+ within hours of dosing. The 500 mg dose in this formula sits within the range most studied for effects on energy metabolism, muscle insulin sensitivity, and physical performance.
NAD+ is the required substrate for sirtuins (SIRT1–SIRT7) the proteins that regulate DNA repair, gene expression, and metabolic adaptation and for PARP enzymes, which use NAD+ to coordinate cellular DNA repair. Maintaining NAD+ levels supports both pathways. Unlike NMN resveratrol combinations, this is a pure NMN supplement designed to address NAD+ availability directly at the source.
