DL-ALA: The Racemic Form

Alpha-lipoic acid has one chiral center, producing two enantiomers: R-ALA (naturally occurring, the form used as a mitochondrial enzyme cofactor) and S-ALA (synthetic). DL-ALA is the 50:50 racemic mixture of both. At 600mg DL-ALA per capsule, the formula provides approximately 300mg of the biologically active R-ALA enantiomer. DL-ALA is more chemically stable and more cost-effective than pure R-ALA and is the most widely studied form in clinical research.

Antioxidant Mechanisms

ALA is reduced to dihydrolipoic acid (DHLA) by lipoamide dehydrogenase. DHLA directly scavenges reactive oxygen species and regenerates the antioxidant network: DHLA reduces dehydroascorbate back to vitamin C, vitamin C reduces the tocopheroxyl radical back to vitamin E, and DHLA reduces oxidized glutathione (GSSG) back to reduced glutathione (GSH). ALA also chelates pro-oxidant metal ions including copper, iron, cadmium, and mercury, reducing their ability to catalyze free radical generation via the Fenton reaction.

Mitochondrial Cofactor and Glucose Metabolism

In its endogenous role, ALA is covalently bound to the E2 subunit of the pyruvate dehydrogenase complex (PDC, which converts pyruvate to acetyl-CoA, linking glycolysis to the citric acid cycle) and the alpha-ketoglutarate dehydrogenase complex (KGDC, a step within the citric acid cycle). At supplemental doses, free ALA activates AMPK (AMP-activated protein kinase), promoting GLUT4 glucose transporter translocation to the cell surface in muscle and adipose tissue and enhancing insulin-stimulated glucose uptake. This glucose-lowering effect is the basis for the diabetes caution and the hypoglycemia stop-use warning.
