This randomized, double blind trial directly compared the efficacy of Vitamin D3 (cholecalciferol) versus Vitamin D2 (ergocalciferol) at equal doses in raising serum 25-hydroxyvitamin D (25(OH)D) levels in healthy adults over 12 weeks. Participants received either 1,000 IU of D3 or 1,000 IU of D2 daily. At 12 weeks, the D3 group showed significantly higher serum 25(OH)D levels compared to the D2 group, with D3 demonstrating approximately 87% greater potency than D2 in raising and maintaining 25(OH)D blood levels. The authors concluded that Vitamin D3 is significantly more effective than D2 at raising serum 25(OH)D the storage and clinically measured form of Vitamin D and recommended D3 as the preferred supplemental form. This finding directly supports the use of cholecalciferol (D3) as the active ingredient in this formula, and the clinical relevance of 25(OH)D blood level monitoring when supplementing at 2,500 IU per day.

This landmark systematic review and meta analysis of 25 randomized controlled trials (total 11,321 participants) examined the protective effect of Vitamin D supplementation against acute respiratory tract infections. Vitamin D supplementation was associated with a statistically significant reduction in the risk of acute respiratory infection across all participants (adjusted OR 0.88, 95% CI 0.81–0.96).
